Endpoint-transparent Multipath Transport with Software-defined Networks
Multipath forwarding consists of using multiple paths simultaneously to
transport data over the network. While most such techniques require endpoint
modifications, we investigate how multipath forwarding can be done inside the
network, transparently to endpoint hosts. With such a network-centric approach,
packet reordering becomes a critical issue as it may cause critical performance
degradation.
We present a Software Defined Network architecture which automatically sets
up multipath forwarding, including solutions for reordering and performance
improvement, both at the sending side through multipath scheduling algorithms,
and the receiver side, by resequencing out-of-order packets in a dedicated
in-network buffer.
We implemented a prototype with commonly available technology and evaluated
it in both emulated and real networks. Our results show consistent throughput
improvements, thanks to the use of aggregated path capacity. We give
comparisons to Multipath TCP, where we show our approach can achieve a similar
performance while offering the advantage of endpoint transparency
Multicomponent Synthesis of Polyphenols and Their In Vitro Evaluation as Potential \u3b2-Amyloid Aggregation Inhibitors
While plant polyphenols possess a variety of biological properties, exploration of chemical
diversity around them is still problematic. Here, an example of application of the Ugi multicomponent
reaction to the combinatorial assembly of artificial, yet \u201cnatural-like\u201d, polyphenols is presented. The
synthesized compounds represent a second-generation library directed to the inhibition of \u3b2-amyloid
protein aggregation. Chiral enantiopure compounds, and polyphenol-\u3b2-lactam hybrids have been
prepared too. The biochemical assays have highlighted the importance of the key pharmacophores in

Genome-wide expression profiling and functional characterization of SCA28 lymphoblastoid cell lines reveal impairment in cell growth and activation of apoptotic pathways
BACKGROUND:
SCA28 is an autosomal dominant ataxia associated with AFG3L2 gene mutations. We performed a whole genome expression profiling using lymphoblastoid cell lines (LCLs) from four SCA28 patients and six unrelated healthy controls matched for sex and age.
METHODS:
Gene expression was evaluated with the Affymetrix GeneChip Human Genome U133A 2.0 Arrays and data were validated by real-time PCR.
RESULTS:
We found 66 genes whose expression was statistically different in SCA28 LCLs, 35 of which were up-regulated and 31 down-regulated. The differentially expressed genes were clustered in five functional categories: (1) regulation of cell proliferation; (2) regulation of programmed cell death; (3) response to oxidative stress; (4) cell adhesion, and (5) chemical homeostasis. To validate these data, we performed functional experiments that proved an impaired SCA28 LCLs growth compared to controls (p\u2009<\u20090.005), an increased number of cells in the G0/G1 phase (p\u2009<\u20090.001), and an increased mortality because of apoptosis (p\u2009<\u20090.05). We also showed that respiratory chain activity and reactive oxygen species levels was not altered, although lipid peroxidation in SCA28 LCLs was increased in basal conditions (p\u2009<\u20090.05). We did not detect mitochondrial DNA large deletions. An increase of TFAM, a crucial protein for mtDNA maintenance, and of DRP1, a key regulator of mitochondrial dynamic mechanism, suggested an alteration of fission/fusion pathways.
CONCLUSIONS:
Whole genome expression profiling, performed on SCA28 LCLs, allowed us to identify five altered functional categories that characterize the SCA28 LCLs phenotype, the first reported in human cells to our knowledge. \ua9 2013 Mancini et al.; licensee BioMed Central Ltd

Musculoskeletal injuries in gastrointestinal endoscopists: a systematic review
Gastrointestinal(GI) endoscopy forms a significant proportion of clinicians' workloads. However, little attention is given to the ergonomic aspects of endoscopy. This systematic review of musculoskeletal pain and/or injuries in GI endoscopists aims to better understand the types of occupational injuries resulting from endoscopic procedures and associated risk factors. Areas covered: Systematic literature search conducted for articles evaluating prevalence, risk factors and mechanism of musculoskeletal pain and/or injuries related to GI endoscopy. In 13 included studies, 39-89% of surveyed endoscopists reported pain and/or injuries related to endoscopy. Common areas of pain were the back(15-57%), neck(9-46%), shoulders(9-19%), elbows(8-15%) and hands/fingers(14-82%). Risk factors included procedure volume, time spent doing endoscopy, cumulative time in practice and endoscopist age. Experimental studies showed that forces and loads placed on endoscopists' bodies during procedures place them at risk of occupational injury. Areas of pain differed between novice and experienced endoscopists implying separate mechanisms of injury. Expert commentary: Comprehensive investigation into the prevalence, types, pathophysiology and methods to minimise endoscopy-related musculoskeletal injuries is vital to ensure the continued efficient provision of endoscopy services in the face of rising demands worldwide. Effects of Sleep Deprivation on Surgeons Dexterity
Sleep deprivation is an ordinary aspect in the global society and its prevalence is increasing. Chronic and acute sleep deprivation have been linked to diabetes and heart diseases as well as depression and enhanced impulsive behaviors. Surgeons are often exposed to long hour on call and few hours of sleep in the previous days. Nevertheless, few studies have focused their attention on the effects of sleep deprivation on surgeons and more specifically on the effects of sleep deprivation on surgical dexterity, often relying on virtual surgical simulators. A better understanding of the consequences of sleep loss on the key surgical skill of dexterity can shed light on the possible risks associated to a sleepy surgeon. HBIM Feeding Open Access Vault Inventory Through GeoDB HUB
This paper describes a methodological workflow starting from a punctual informative Historical Building Information Models (HBIM) - derived from the geometric analysis of the construction technology of vaulted systems - used to feed an open access Geospatial Data Base based on a Virtual Hub technology. Vaulted systems, which are characterized by a variety of solutions developed across time and space, and whose knowledge is obtained during the analysis and diagnostic phases planned for preservation purposes, are mostly unknown to the public and collected information risks to be missed. This way, the chain of knowledge transfer is interrupted both for more informed future sustainable interventions and touristic purposes. The potentials offered by the adoption of a Pan European Virtual Hub to manage open data with semantic, spatial and temporal sub-setting allows to reconstruct a new framework of the construction techniques widespread across the world, permanencies and mutations to the common typologies with the contribution of skilled workers moving during the centuries across Europe. The case study of Palazzo Magio in Cremona with a rich abacus of vaults apparently belonging to a simple ‘cloister’ typology is described from the HBIM generation to data publication in the GeoDB HUB. Different Level of Detail, Geometry and Information, acquired in other Cremona Palaces, are feeding the DB and its vocabulary in a bottom-up process. An Autoregulatory Loop Directs the Tissue-Specific Expression of p63 through a Long-Range Evolutionarily Conserved Enhancer
p63, a p53 family member, is essential for the development of various stratified epithelia and is one of the earliest markers of many ectodermal structures, including the epidermis, oral mucosa, apical ectodermal ridge, and mammary gland. Genetic regulatory mechanisms controlling p63 spatial expression during development have not yet been defined. Using a genomic approach, we identified an evolutionarily conserved cis-regulatory element, located 160 kb downstream of the first p63 exon, which functions as a keratinocyte-specific enhancer and is sufficient to recapitulate expression of the endogenous gene during mouse embryogenesis. Dissection of the p63 enhancer activity revealed a positive autoregulatory loop in which the p63 proteins directly bind to and are essential regulators of the enhancer. Accordingly, transactivating p63 isoforms induce endogenous p63 expression in cells that do not normally express this gene, whereas dominant negative isoforms suppress p63 expression in keratinocytes. In addition the transcription factor AP-2 also binds to the enhancer and cooperates with p63 to induce its activity. These results demonstrate that a long-range autoregulatory loop is involved in the regulation of p63 expression during embryonic development and in adult cells
